ABSTRACT

OBJECTIVE@#To observe the effect of acupuncture on visual acuity, intraocular pressure, visual field, retinal and choroidal thickness on optic disc and macular area in patients with optic atrophy.@*METHODS@#A total of 33 patients with optic atrophy were treated with acupuncture. Acupuncture was given at Chengqi (ST 1), Shangjingming (Extra), Qiuhou (EX-HN 7) and Fengchi (GB 20) etc., 30 min each time, once a day, for 14 days. The visual acuity, intraocular pressure, visual field indexes (mean deviation [MD], pattern standard deviation [PSD] and visual field index [VFI]), optic disc retinal nerve fiber layer thickness, macular retinal thickness and choroidal thickness of optic disc and sub-foveal were compared before and after treatment.@*RESULTS@#Compared before treatment, the visual acuity was increased (P<0.05), the MD value was decreased (P<0.05), the thickness of nerve fiber layer on the upper temporal side of optic disc was thinner (P<0.05), and the choroidal thickness of average, nasal side and lower temporal side of optic disc was increased (P<0.05). There was significant correlation between visual field MD and retinal nerve fiber layer thickness in different quadrants before and after treatment (P<0.01).@*CONCLUSION@#Acupuncture could improve visual acuity, increase choroidal thickness in part of optic disc area in patients with optic atrophy.

ABSTRACT

Objective::To study the chemical structure and morphological features of a polysaccharide BHP-1 isolated and purified from the bulbs of Lilium davidii var. unicolor. Method::Thermogravimetry (TG) and scanning electron microscopy (SEC) was used to determine the thermal characteristics and the surface morphology changes of the polysaccharide BHP-1, respectively. The chemical structure of BHP-1 was analyzed by gas chromatography-mass spectrometer (GC-MS), partial acid hydrolysis, sodium periodate oxidation-Smith degradation and nuclear magnetic resonance (NMR). Result::The results showed that the backbone of the polysaccharide BHP-1 mainly contained α-(1→4)-linked D-glucopyranosyl and β-(1→4)-linked D-mannopyanosyl, and the branches were probably linked at the O-2 and/or O-3 of the mannosyl and glucosyl residues, with T-α-D-glucopyranosyl as a terminal structure. It was a natural mannoglucose containing a small amount of O-acetyl group. BHP-1 began to degrade at 220 °C, and the degradation ended at 520 ℃, indicating a good thermal stability, smooth surface and a large number of depressions. The depressions were formed by closely interlaced and sunken sheet layers with irregular holes. Conclusion::BHP-1 was a natural mannoglucose with a good thermal stability, smooth surface, a large number of depressions and irregular holes, and contained a small amount of O-acetyl group. Its chemical structure was reported for the first time.

ABSTRACT

AIM To explore the effects of Bushen Huoxue Recipe (Fluoritum,Psoraleae Fructus,Cuscutae Semen,etc.) on the protein expressions of transforming growth factor (TGF)-β1,TGF-βRⅡ and Smad2/3 in follicle wall granulosa cells in mice with autoimmune premature ovarian failure (POF).METHODS Balb/c female mice were subcutaneously injected with mouse zona pellucida 3 at multiple points to establish autoimmune POF model.POF mice were divided into model group,positive group (progynova),low-,middle-and high-dose of Bushen Huoxue Recipe groups.After intervention for 30 days,ovarian tissue was stained by hematoxylin-eosin (HE),and the protein expressions of TGF-β1,TGF-βRⅡ and Smad2/3 in follicle wall granulosa cells and ovarian tissue were detected by immunohistochemistry and Western blot,respectively.RESULTS Compared with the model group,the number of mature follicles in the Bushen Huoxue Recipe groups and the positive group was obviously increased,together with the decreased number of atretic follicles.The protein expressions of TGF-β1,TGF-βRⅡ and Smad2/3 in follicle wall granulosa cells and ovarian tissue in the Bushen Huoxue Recipe groups and the positive group were much higher than those in the model group (P < 0.05).CONCLUSION Bushen Huoxue Recipe can improve ovarian function by up-regulating the protein expressions of TGF-β1,TGF-βRⅡ and Smad2/3 in granulosa cells.

ABSTRACT

An electrochemical sensor was fabricated for determination of cholesterol ( ChO ) based on the molecularly imprinting and signal amplification of graphene. The sensor was prepared by electropolymerizing phenol on the surface of graphene modified glassy carbon electrode with ChO as template. The structure, properties and molecular imprinting effect of the imprinted membrane were studied by SEM, CV and DPV. The results showed that the sensor had good selectivity and high sensitivity to cholesterol. The calibration graph for the determination of cholesterol was linear in the range of 8. 0 × 10-8-2. 0 × 10-4 mol/L, with the detection limit (S/N=3) of 5. 6×10-8 mol/L. The sensor was applied to the determination of cholesterol in human serum samples with satisfactory results.

Objective To analyze the clinical application of TCM decoction pieces for heat clearing in the First Affiliated Hospital of Xi'an Jiaotong University; To promote rational administration of TCM decoction pieces, and guarantee patients' medication safety. Methods By using Microsoft Office Excel 2013 software, use frequencies, classifications of pharmacological actions and doses of TCM decoction pieces for heat clearing (DHC) from 2013 to 2015 of the First Affiliated Hospital of Xi'an Jiaotong University were analyzed. Results In our hospital, from 2013 to 2015, in top 15 DHC, the amounts of clearing away heat and toxin were more than 40%, and which of the average doses exceeded the upper limit of pharmacopoeia dosage, were more than 30%. The part that overdoes frequencies greater than 30% were 11.0% of the total, nevertheless, the part that not exceeded was 39.0% of the total. Conclusion The use of medicine for heat clearing should be carefully dialectical, paying attention to the rationality of medication. The dose should be careful and effectively regulated.

ABSTRACT

BACKGROUND: Transplantation of human umbilical cord mesenchymal stem cells can effectively promote the recovery of neurological function in animal models of ischemic brain injury. However, it is unclear on the mechanisms underlying the therapeutic actions of human umbilical cord mesenchymal stem cells and the optimal transplantation way. Bioluminescence imaging has been widely used in the field of stem cell transplantation for real-time dynamic monitoring of the proliferation, migration and survival of transplanted stem cells. OBJECTIVE: To review the studies of human umbilical cord mesenchymal stem cells in the treatment of ischemic brain injury and the recent research progress of bioluminescence imaging in the stem cell transplantation for the treatment of ischemic brain injury. METHODS: The key words were "human umbilical cord mesenchymal stem cells, brain ischemia, bioluminescence imaging" in English and Chinese, respectively. The first author retrieved PubMed database, Chinese Journal Full-text Database for relevant articles published from January 2011 to December 2016. Literatures with repetitive content and poor relationship were excluded. A total of 98 literatures were initially retrieved, and finally 38 articles met the inclusion criteria. RESULTS AND CONCLUSION: Human umbilical cord mesenchymal stem cells can be transplanted via the vein, artery, lumbar puncture, stereotactic operation, intraventricular approach in the treatment of ischemic brain injury. They have their own merits and demerits, which mainly focus on stem cell migration range, survival rate, safety and so on. It has been found that bioluminescence imaging technology for the living is characterized by real-time dynamic monitoring, high sensitivity, high-time resolution and easy operation, which can be used to monitor the migration, proliferation and survival of transplanted stem cells in vivo. Therefore, further investigation on the mechanism of human umbilical cord mesenchymal stem cells that are labeled by bioluminescence imaging technology and selection of the optimal transplantation approach is warranted.

ABSTRACT

Objective To observe the clinical efficacy of Wenshen Xiaochuan Paste Recipe (WXPR) in preventing bronchial asthma, and to explore the mechanism. Methods Eighty patients were randomly divided into treatment group and control group. Each group had 40 patients. The control group was treated by conventional western medicine such as inhalation of steroid, and the treatment group was given oral use of WXPR based on the treatment for the control group. Treatment for both groups covered 60 days, and the follow-up lasted 10 months. The clinical outcomes included the clinical efficacy and immune indexes of peripheral blood T lymphocyte subsets CD3+, CD4+, CD8+and CD4+/CD8+ratio, and the serum IgE level. Results (1) After treatment and 12 months later, the scores of asthma control test (ACT ) in the two groups were increased obviously as compared with those before treatment, and there were statistically significant differences (P0.05) , but were significant ( P0.05). The improvement of immune indexes in the treatment group was superior to that in the control group ( P<0.05). Conclusion WXPR has good clinical efficacy in preventing and treating bronchial asthma. The mechanism is related with the inhibition of airway inflammation and improvement of immune function probably through regulating T lymphocyte subsets and lowering serum IgE level in asthma patients.

ABSTRACT

<p><b>OBJECTIVE</b>To optimize the evaluation level of active motor threshold and the functional domain of upper limbs of stroke-specific quality of life (SS-QOL) scale with Rasch analysis.</p><p><b>METHODS</b>Sixty patients with acute ischemic stroke that were in accord with research criterid were randomly divided into a test group (30 cases) and a control group (30 cases). Acupuncture treatment and routine western medicine were applied on the test group, and single treatment of routine western medicine was applied on the control group. Selected acupoints were MS 5, Fengchi (GB 20), Hegu (LI 4), etc. Active motor threshold and the functional domain of upper limbs of SS-QOL were self-tested by patients after one treating course. The characteristics of the above mentioned items were tested with Rasch model.</p><p><b>RESULTS</b>The statistical result on fitness of active motor threshold and the functional domain of upper limbs of SS-QOL showed that every reference of samples and items wosin accord with the Rasch model and has well inner reliability and validity. The Infit and Outfit MnSq values of active motor threshold and the functional domain of upper limbs of SS-QOL are basically between 0.5 and 1.5.</p><p><b>CONCLUSION</b>The application of Rasch analysis on the assessment of patient reported outcome (PRO) has optimized the PRO scale (the activity and upper limb function domain of SS-QOL scale) and enhanced evaluation level of active motor threshold and the functional domain of upper limbs of SS-QOL scale.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of TSP02, a triterpenoid saponin compound from Ardisia japonica, on proliferation and metastasis of in vitro induced human hepatoma HepG2 cells and its molecular mechanism.</p><p><b>METHOD</b>MTT assay was performed to detect the inhibitory effect of TSP02 of different concentrations on proliferation of human hepatoma HepG2 and normal human hepatic cell HL-7702 cells. The changes in cell cycle and apoptotic of processed HepG2 and HL-7702 cells were detected by using flow cytometry. The effect of TSP02 on expression levels of CDK1, 2, 4, apoptosis-related protein Caspase-8, metastasis-related TGF-beta 1 and E-cadherin was measured by western blot. Wound-healing assay and transwell assay were performed to detect the changes in the metastasis of TSPO2-processed HepG2.</p><p><b>RESULT</b>TSP02 significantly inhibited the proliferation of HepG2 cells, with notable time dependence and concentration dependence, but without remarkable effect on normal human liver HL-7702 cells. Compared with the control group, TSP02 processed for 24 h could eliminate HepG2 cells in S stage, significantly increase the cell apoptotic rate. Furthermore, TSP02 was capable of down-regulating the expression of multiple CDK1, 2, 4, and TGF-beta1, and up-regulating the expression and activity of Caspase-8, without significant effect on cycle and apoptotic rate of normal human liver HL-7702 cells. Additionally, TSP02 caused metastasis and invasiveness HepG2 cells, while down-regulating liver cancer invasiveness-related TGF-beta 1 and E-cadherin.</p><p><b>CONCLUSION</b>TSP02 selectively promotes apoptosis of liver cancer cell HepG2, and inhibits its metastasis and invasiveness. TSP02's in vitro antineoplastic activity is related to the changes in cycle and apoptosis proteins, and the regulation in the expression of invasiveness-related TGF-beta 1 and E-cadherin.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the syndrome evolution law of Chinese medicine (CM) in the patients with gastric mucosal dysplasia.</p><p><b>METHODS</b>Three hundred and twenty four gastric mucosal dysplasia patients with deficiency and excess correlation syndromes were enrolled by a multi-center collaboration for two years' clinical follow-up to detect the levels of tumor supplied group of factors (TSGF) and carcino-embryonic antigen (CEA).</p><p><b>RESULTS</b>Among the 324 cases, 29 cases turned cancer in the two years, and the canceration rate was 9.0%. The three syndromes with higher canceration rate were the damp-heat accumulating Wei syndrome concurring or combining with asthenia-cold in Pi and Wei syndrome for 16.7%; stagnation in Wei collaterals syndrome concurring or combining with asthenia of both qi and yin syndrome for 13.2%; stagnation of Gan and Wei qi syndrome concurring or combining with asthenia-cold in Pi and Wei syndrome for 8.0%, respectively. Among the three syndromes, the highest level of TSGF occurred in the former two syndromes. In the half year before carcinogenesis, the syndromes of the patients took on deficiency and excess concurrent syndromes, and the deficiency syndromes involving the qi and blood deficiency syndrome and the Shen deficiency syndrome accounting for 48.0%.</p><p><b>CONCLUSIONS</b>Gastric mucosal dyspalsia canceration syndromes took on the polymorphism of excess and deficiency concurrent syndromes and had the characteristics of deficiency syndromes involving qi and blood deficiency syndrome and Shen-yin-yang deficiency syndrome.</p>

ABSTRACT

Objective To assess the capability of monochromatic energy images of Gemstone spectral imaging(GSI) in reducing artifacts caused by metallic implants. Methods Twelve subjects with metallic implants underwent GSI (Discovery CT750 HD, GE Healthcare, Milwaukee ). The metallic orthopedic implants included 3 patients of dentures, 2 patients of cervical spinal vertebraplasty, one clavicle fracture fixation, one lumbar spinal vertehraplasty, 3 patients of artificial femoral head, one iliac fracture fixation and one tibial fracture fixation. GSI was performed by using a single source ultra-fast dual energy X-ray switching (80 kVp and 140 kVp). Following GSI scanning, thin slice images were reconstructed into 1.25 mm slice thickness. The monochromatic energy images were set to the same window width and level (window width 1500 HU,window level 500 HU). The artifact indexes (AI) at different kiloelectronvolts (keV) images were measured and compared. 3D reconstruction was performed using images with minimal AI. Result The artifacts index on monochromatic energy images varied with the change of keV. Of the images from 12 subjects, the maximal AI ranged between 145-225 at 40 keV, and minimal AI ranged between 15-90 at the 95-140 keV. The artifacts are clearly visible on polychromatic energy images and the artifacts are reduced markedly on the monochromatic energy images with minimal AI. Conclusion The artifacts caused by metallic implants can be reduced significantly by GSI with high keV monochromatic energy images.

ABSTRACT

AIM:To explore the potentially beneficial clinical effects of traditional Chinese medicine(TCM) combined with photocoagulation for diabetic retinopathy(DR).METHODS:Chinese patients with DR were divided into two groups. A joint treatment group received both the TCM ziyinliangxuesanyutang and photocoagulation, while a control group received only photocoagulation laser treatment. Visual acuity tests, visual field retinal sensitivity tests, and fundus fluorescein angiography (to measure neovascular regression) were performed. Vision was compared between the two groups 1 month, 6, and 12 months after treatment.RESULTS:Twelve months after treatment, the recovery of visual acuity (62.3% vs 43.1%, P=0.037) and retinal sensitivity \[17.0±3.7 decibels (dB) vs 14.9±3.7dB, P=0.002\] as well as neovascular regression (67.2% vs 48.3%, P=0.036) in the joint treatment group were all significantly greater than that of the control group.CONCLUSION:Compared with laser treatment alone, the joint application of TCM and photocoagulation is shown to be more effective than DR treatment method.

ABSTRACT

Gene mutation of Candida albicans is one of the main causes for azole drug resistance. Different types of variation play different roles in promoting the process of drug resistance. ERG series of gene mutations primarily affect the ergosterol synthesis pathway. When the regulatory factors TAC1 for CDR1 gene and Mrr1 for MDR1 gene generate mutations, the expression level of drug efflux pump protein in Candida albicans may be changed. In addition, gene copy number variation is also gaining attention. Therefore, the research of mutation resistance-associated genes has a positive meaning to explore the mechanism of drug resistance in Candida albicans.

ABSTRACT

<p><b>OBJECTIVE</b>To study the expression of cell cycle related factor sonic hedgehog (SHH) in autogenous vein graft and its relation with neointima formation.</p><p><b>METHODS</b>Autogenous vein graft model were established in 24 male Wistar rats of 8 weeks old and 140 g weight, by transplanting the left jugular vein to intra renal abdominal aorta with microsurgical technique. Graft veins were harvested at 14 d and 28 d after transplantation. The immunohistochemistry and Western blot were used to detect the SHH and PCNA expression in the vein graft. At the same time SHH mRNA was measured by quantitative real-time PCR. The opposite normal veins served as control.</p><p><b>RESULTS</b>Histological staining showed that the percent of SHH+ cells was only (2.0 +/- 0.5)% in the normal vein, but was much more in the vein graft after surgery, as (39.4 +/- 0.4)% and (63.0 +/- 0.3)% respectively (P < 0.01). The expression of SHH and PCNA were both elevated in the vein graft. There was a positive correlation between them which indicated by Western blot (r = 0.808, P < 0.01). The SHH mRNA content also increased in vein graft to 9.5 and 23.8 folds of that in control.</p><p><b>CONCLUSION</b>SHH is upregulated in autogenous vein grafts and may correlated with the proliferation of vascular smooth muscle cells.</p>

ABSTRACT

This study was aimed to evaluate the reversed effects of cyclosporin A (CsA) on multidrug resistance (MDR) of human leukemic cell line HL-60/ADM, and to investigate the relationship of the oxygen free radical content between HL-60/ADM cells and the reversed HL-60/ADM cells (HL-60/ADM + CsA). The cytotoxicity and the reversed effects of CsA on multidrug resistance of human leukemic cell line HL-60/ADM were studied by MTT, flow cytometry (FCM) and immunohistochemical assay; the oxygen free radical for HL-60/ADM and HL-60/ADM + CsA cell lines were detected by colorimetric method. The results showed that the CsA less than 4 microg/ml had no significant cytotoxicity on HL-60/ADM, while the cytotoxicity was rised with CsA concentration increasing; And CsA (4 microg/ml) combined with ADM (1 microg/ml) could obviously restrain the growth of HL-60/ADM cells (p < 0.001). The P-gp expression of HL-60/ADM decreased obviously after exposure to CsA (4 microg/ml) for 72 hours, at the same cell conditions, MDA concentration of the reversed groups (HL-60/ADM + CsA cells) was higher than that of the control groups (HL-60/ADM cells) (p < 0.05), while the levels of SOD and GSH in the reversed groups were significantly lower than that in control groups (p < 0.001). It is concluded that MDR of HL-60/ADM can be reversed effectively by low dose of CsA, the level of oxygen free radical increases and the activity of antioxidants decreases in the reversed cells. Oxygen free radicals may be involved in this reverse process, which thereby lead to the cell death.

ABSTRACT

Polyamides, containing N-methylpyrrole (Py) and N-methyl-imidazole (Im) amino acids, are synthetic oligomers programmed to read the DNA double helix in the minor groove with high affinities and sequence specificities resulting in modulation of gene expression. They are cell permeable, stable and have no cytotoxicity, which provide a promising tool of gene regulation. We describe here recent advances in the field of DNA binding polyamides, including pairing rules, specifities and affinities to DNA, synthesis methods, cellular and nuclear uptake properties, gene regulation and effectiveness in vivo. The potential problems and difficulties in future research are also discussed.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the safety and efficacy of sonographically guided percutaneous intralesional sclerotherapy for peripheral venous malformations.</p><p><b>METHODS</b>From March 2004 to October 2007, 32 patients with venous malformations of soft tissues were treated with sonographically guided intralesional sclerotherapy. The malformed venous space was identified intraoperatively by duplex scanning, the gauge 7 needle was inserted into this venous space under real-time ultrasound visualization, and sclerosants were infused in the space gently. Absolute alcohol and bleomycin acted as sclerosants here, combined the two (for type I malformation) or bleomycin only (for type II malformation). The treatment could be repeated if the lesion was not cured in 3 weeks. All patients were followed up for 6 months to 2 years.</p><p><b>RESULTS</b>Of the group, each patient received 1 - 6 times of the therapy (mean, 3 times). Twenty-seven patients achieved a complete response, and 5 achieved partial response. No major complications occurred.</p><p><b>CONCLUSIONS</b>Sonographically guided percutaneous intralesional sclerotherapy for peripheral venous malformation is a simple, effective, safe therapy with minimal invasion, lower morbidity rate, and can be repeated.</p>
